A Study to Identify Late-Occurring Complications in Childhood Cancer Survivors Rochester, Minn. The purpose of this trial is to identify cancer survivors who are at increased risk of developing late-occurring complications after undergoing treatment for childhood cancer. A patient's genes may affect the risk of developing complications, such as congestive heart failure, heart attack, stroke, and second cancer, years after undergoing cancer treatment. Genetic studies may help doctors identify survivors of childhood cancer who are more likely to develop late complications. ACT-MBC: A Study of Circulating Tumor Cells (CTCs) in Metastatic Breast Cancer (MBC) (ACT-MBC) Mankato, Minn., Rochester, Minn., Eau Claire, Wis., Albert Lea, Minn. Collection of blood to track serial circulating tumor cells (CTCs) in subjects with metastatic breast cancer (MBC). Study will also collect data from investigators are Mayo Clinic and the Mayo Clinic Health Systems to determine effectiveness of the proposed process. Chemotherapy Alone or Chemotherapy Plus Radiation Therapy in Treating Patients with Locally Advanced Rectal Cancer Undergoing Surgery Rochester, Minn., Scottsdale/Phoenix, Ariz. The standard treatment for locally advanced rectal cancer involves chemotherapy and radiation, known as 5FUCMT, (the chemotherapy drugs 5-fluorouracil/capecitabine and radiation therapy) prior to surgery. Although radiation therapy to the pelvis has been a standard and important part of treatment for rectal cancer and has been shown to decrease the risk of the cancer coming back in the same area in the pelvis, some patients experience undesirable side effects from the radiation and there have been important advances in chemotherapy, surgery, and radiation which may be of benefit. The purpose of this study is to compare the effects, both good and bad, of the standard treatment of chemotherapy and radiation to chemotherapy using a combination regimen known as FOLFOX, (the drugs 5-fluorouracil (5-FU), oxaliplatin and leucovorin) and selective use of the standard treatment, depending on response to the FOLFOX. The drugs in the FOLFOX regimen are all FDA (Food and Drug Administration) approved and have been used routinely to treat patients with advanced colorectal cancer. Tissue Collection for Patients with Cancers other than Hepatobilairy Cancers with Metastasis to the Liver Rochester, Minn. The goal of this study is to develop a resource (bank) of biospecimens (blood, tissue) from patients having cancers that metastasis to the liver. The purpose of the resource bank is to facilitate studies on the pathogenesis of cancers metastasizing to the liver and also in order to assess the ability of new tests to detect early cancer in the cohort of patients undergoing routine surveillance for cancer. The biospecimens will include cultures of tumor cells and tumor-associated non-malignant cells, as well as xenografts established from freshly isolated metastatic cancers in immunodeficient mice. We plan to administer a risk factor questionnaire to facilitate identification of risk factors for development of metastatic liver cancer. The study will also determine survival of patients from time of diagnosis. A Study of ONO-7475 in Patients With Acute Leukemias Jacksonville, Fla. The purpose of this study is to assess the safety and tolerability of ONO-7475 monotherapy in patients with relapsed or refractory acute myeloid leukemia or relapsed or refractory myelodysplastic syndromes, and to assess the safety, tolerability, and preliminary effectiveness of the combination of ONO-7475 and venetoclax in patients with relapsed or refractory acute myeloid leukemia. A Study to Evaluate Rectal Cancer with Diffusion-Weighted and Dynamic Contrast Enhanced Perfusion-Weighted Magnetic Resonance Imaging Jacksonville, Fla. The purpose of this study is to analyze the apparent diffusion coefficient (ADC) from Diffusion-Weighted (DWI) in patients before, during the second week of and after preoperative chemoradiation therapy for rectal cancer. A Registry to Collect Patient and Family DNA and Information for Anyone who has had Multiplex Testing and Gene Mutation Results Rochester, Minn. The purpose of this study is to collect patient and family DNA and disease data to learn more about how changes in certain genes may be linked to cancer. Some people with cancer may have aquired their disease because they inherited an abnormal (mutated) gene. The researchers of this study want to better understand the risks that are linked to genetic changes in these less well-studied genes. By understanding these risks, it is believed that doctors will be able to give better advice to families with mutations in these genes. A Study to Evaluate the Surgical Removal of Residual Brain Tumors Prior to Recurrence Rochester, Minn. The purpose of this study is to evaluate the safety and feasibility of surgical resection for pre-recurrent brain tumors. Eligible patients with surgically accessible latent tumors desiring surgical resection will be enrolled to prospectively track short- and long-term outcomes. Safety will be evaluated by quantifying rates of surgical morbidity as compared to patients undergoing RT after surgery, or no surgery for similar latent disease. Variables evaluated will include postoperative complications including death within 30 days, wound infection, length of hospital stay, and readmission rates. Reduced Craniospinal Radiation Therapy and Chemotherapy in Treating Younger Patients With Newly Diagnosed WNT-Driven Medulloblastoma Rochester, Minn. This phase II trial studies how well reduced doses of radiation therapy to the brain and spine (craniospinal) and chemotherapy work in treating patients with newly diagnosed type of brain tumor called WNT)/Wingless (WNT)-driven medulloblastoma. Recent studies using chemotherapy and radiation therapy have been shown to be effective in treating patients with WNT-driven medulloblastoma. However, there is a concern about the late side effects of treatment, such as learning difficulties, lower amounts of hormones, or other problems in performing daily activities. Radiotherapy uses high-energy radiation from x-rays to kill cancer cells and shrink tumors. Drugs used in chemotherapy, such as cisplatin, vincristine sulfate, cyclophosphamide and lomustine, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving reduced craniospinal radiation therapy and chemotherapy may kill tumor cells and may also reduce the late side effects of treatment. Niraparib + TSR042 In BRCA Mutated Breast Cancer Rochester, Minn. The purpose of this study is to evaluate pre-operative therapy that is specifically targeted for breast cancer in individuals with BRCA mutations.
