The study purpose is to understand patients’ with the diagnosis of Diabetes Mellitus type 1 or 2 perception of the care they receive in the Diabetes clinic or Diabetes technology clinic at Mayo Clinic and to explore and to identify the healthcare system components patients consider important to be part of the comprehensive regenerative care in the clinical setting. However, before we can implement structural changes or design interventions to promote comprehensive regenerative care in clinical practice, we first need to characterize those regenerative practices occurring today, patients expectations, perceptions and experiences about comprehensive regenerative care and determine the factors that promote and hinder their occurrence. Therefore, as exploratory work to inform the development of a comprehensive regenerative care clinic concept, we propose a qualitative study to explore patient perceptions of what it means to experience comprehensive regenerative care in the context of a clinical encounter. We hypothesize that the findings from this study will improve the care of patients with diabetes based on the concept of comprehensive regenerative care. The findings from this study may also lead to better care plans that are congruent with patient goals and context, therefore leading to improvements in outcomes that matter to these patients. Insulin Regulation of Lipolysis and Lipolysis Proteins Rochester, Minn. The purpose of this study is to determine whether impaired insulin-induced suppression of lipolysis (as measured by IC50) is related to lipolysis proteins in groups of volunteers known to vary widely with regards to abdominal adipocyte size and regulation of adipose tissue lipolysis, and whether the improved insulin regulation of lipolysis resulting from treatment with the PPARγ agonist pioglitazone, with or without weight loss, can be linked to specific changes in sets of PPARγ-responsive adipocyte lipolysis proteins in UBO adults. A Study to Test How Muscle Tissue Differs Between People Rochester, Minn. The purpose of this study is to look for ways to test how muscle tissue differs between people. A Study to Evaluate the Safety and Effectiveness of Crinecerfont in Adults with Classic Congenital Adrenal Hyperplasia Rochester, Minn. The purpose of this study is to evaluate the effectiveness of NBI-74788 (100 mg twice daily [bid]), compared with placebo, in reducing daily glucocorticoid dosage while maintaining adrenal androgen control, in reducing adrenal steroid levels following an initial 4-week treatment period, on clinical endpoints associated with supraphysiologic (greater than normal) glucocorticoid dosing, to evaluate plasma concentrations of NBI-74788 and metabolites, and to assess the safety and tolerability of NBI-74788. A Study to Evaluate Cellular Senescence with Senolytics to Improve Skeletal Health in Older Humans Rochester, Minn. The purpose of this study is to test whether intermittent senolytic therapy reduces markers of biological age and improves bone turnover markers and skeletal parameters in older postmenopausal women. A Study of Selpercatinib to Treat Participants with RET-Mutant Medullary Thyroid Cancer Rochester, Minn., Scottsdale/Phoenix, Ariz., Jacksonville, Fla. The purpose of this study is to evaluate the safety and effectiveness of Selpercatinib, compared to a standard treatment, in participants with rearranged during transfection (RET)-mutant medullary thyroid cancer (MTC) that cannot be removed by surgery or has spread to other parts of the body. Participants who are assigned to the standard treatment and discontinue due to progressive disease have the option to potentially crossover to selpercatinib. Pediatric Obesity Weight Evaluation Registry (POWER) Study Rochester, Minn. The main objective of this study is to establish a national pediatric obesity registry known as POWER (Pediatric Obesity Weight Evaluation Registry). This registry will contain clinical data from individual comprehensive pediatric weight management programs around the United States for overweight and obese youth. A Study to Predict Recovery from Cushing Syndrome Rochester, Minn. The primary purpose of this study is to determine pre-surgical and post-surgical predictors of the duration and severity of glucocorticoid withdrawal in patients undergoing a curative surgery for cortisol excess. A Study to Evaluate the Impact of Nutrition Education and Access to Nutritious Foods on Diabetes Control Mankato, Minn. The purpose of this study is to evaluate whether or not a 6 month supply (1 meal//day) of healthy food choices readily available in the patient's home and self management training including understanding of how foods impact diabetes, improved food choices and how to prepare those foods, improve glucose control. In addition, it will evaluate whether or not there will be lasting behavior change modification after the program. A Study to Establish an Obesity-Related Phenotype Registry Rochester, Minn. The purpose of this study is to create a registry of obese patients who have been phenotyped to understand the heterogeneity of obesity.
